    The spatial organisation of DNA in the nucleus starts with Lamina Associated Domains (LADs) tethered to the nuclear envelope, followed by knots of A compartments that house tighter knots of Topologically Associated Domains (TADs) and chromatin loops. TADs and loops are further coloured by different flavours of histone modifications that quantitatively regulate gene networks. Disease-inducing conditions can cause critical imbalances within this landscape, the effects of which may trickle down to gene expression. An established genetic code allows us to understand of the consequences of direct mutational changes on coding DNA, however, a ‘standard model’ linking epigenomic cues and 3D genome-organisation to gene expression has been very difficult to create. My research ambition has been to understand and delineate a better global model of gene expression.
