The average adult now spends 7+ hours per day on screens, driving an epidemic of dry eye, digital eye strain, and blue light-induced oxidative damage to the macula. We are formulating a daily softgel targeting this population, combining lutein (10mg), zeaxanthin (2mg), astaxanthin (6mg), omega-3 DHA/EPA (250mg), and vitamin A (400µg) in an oil-based suspension for optimal bioavailability. The product will be sold direct-to-consumer in the USA, UK and EU as a food supplement (not a medicine). We are seeking an independent professional opinion on the formulation before approaching contract manufacturers. We are not yet committed to any manufacturer or ingredient supplier. Proposed Formulation (Per Softgel) Lutein 10 mg Free-form or esterified (from Tagetes erecta marigold extract) Zeaxanthin 2 mg From marigold extract or synthetic Astaxanthin 6 mg Natural (from Haematococcus pluvialis) Omega-3 (DHA + EPA combined) 250 mg Algal oil (preferred) or fish oil rTG/EE Vitamin A 400 µg RE As retinyl palmitate or retinyl acetate Carrier oil TBD Sunflower or safflower (to be confirmed) Antioxidant system TBD To protect astaxanthin and omega-3 from oxidation Target softgel shell: Oil-based suspension in a bovine gelatin or vegan (HPMC/starch) shell. Shell colour preference is matte black — we would like your view on whether this is achievable without compromising the shell integrity or regulatory status. Questions for Your Review 1. Formulation Viability • Is this combination of ingredients physically compatible in a single oil-based softgel? Are there any known interactions between astaxanthin, lutein/zeaxanthin, and omega-3 in a lipid matrix that could affect potency or stability? • Is 250mg of omega-3 (DHA+EPA) sufficient to deliver a meaningful clinical effect for dry eye and tear film support, or would you recommend increasing the dose? What is the practical upper limit for a single softgel of standard size (e.g., 20 oblong)? • Is 6mg astaxanthin an appropriate dose for digital eye strain? The EU Novel Food authorisation for astaxanthin permits up to 8mg/day — is there a clinical or safety reason to stay below this ceiling, or should we go to 8mg? • Should meso-zeaxanthin be included alongside lutein and zeaxanthin? The UKNHCC reviewed a claim for the 10:2:10 (lutein:zeaxanthin:meso-zeaxanthin) combination but concluded evidence was insufficient for a specific health claim. Does the clinical evidence support including it regardless, and what would it add to the formulation? 2. Ingredient Sourcing & Quality • What are the leading branded ingredient suppliers you would recommend for each active, and why? We are particularly interested in: • Lutein/Zeaxanthin: e.g., FloraGLO® (Kemin), Lutemax® 2020 (OmniActive), or generic marigold extract • Astaxanthin: e.g., AstaREAL® (Fuji Chemical), BioAstin® (Cyanotech), or generic H. pluvialis extract • Omega-3 (algal): e.g., life'sDHA™ (DSM), DMAX® (LUS Health), or generic algal oil • Are branded ingredients meaningfully superior in terms of bioavailability and purity, or is the premium primarily a marketing advantage? What would you recommend for a brand at this stage? • For vitamin A, is retinyl palmitate or retinyl acetate more appropriate in an oil-based softgel matrix? 3. Stability & Oxidation • Astaxanthin and omega-3 are both highly susceptible to oxidative degradation. What antioxidant system would you recommend to protect both actives throughout a 24-month shelf life? (e.g., mixed tocopherols, rosemary extract, ascorbyl palmitate — or a combination?) • What is the recommended packaging specification to maximise stability — amber vs. opaque bottle, nitrogen flushing, desiccant inclusion? • Are there any known stability incompatibilities between astaxanthin and the other oil-soluble actives (lutein, vitamin A) in a shared lipid carrier? • What stability testing protocol would you recommend, and at what point in the development process should it be initiated? 4. Regulatory & Claims • Under UK/EU food supplement regulations, are all five ingredients at the proposed doses permissible without any novel food authorisation or notification requirement? How about USA? • The only EFSA-approved health claim we are currently planning to use is "Vitamin A contributes to the maintenance of normal vision." Are there any other approved claims in the GB, US or EU register that could legitimately be used for any of the other ingredients at these doses? • Are there any labelling requirements specific to astaxanthin (e.g., mandatory warnings) that we should be aware of? 5. Format & Manufacturability • Is a single softgel the right format for this combination, or would you recommend splitting into two capsules per day to accommodate the fill volume and protect ingredient stability? • What is your view on vegan (HPMC or starch-based) softgel shells for this formulation? Are there any compatibility issues with the oil-based fill or specific actives? • Can a softgel shell be reliably produced in a dark/black colour using approved food-grade colorants (e.g., activated charcoal, iron oxides)? Does the colorant affect the shell's permeability or the product's regulatory status? Deliverables Requested We would welcome your feedback in the form of a short written report covering: 1. An assessment of the formulation's viability and any recommended adjustments to doses or ingredient forms 2. Ingredient sourcing recommendations (branded vs. generic) with rationale 3. Recommended antioxidant and packaging strategy for stability 4. Any regulatory red flags or labelling requirements we should address before going to manufacture 5. A recommended stability testing protocol
